Комплекс морфологічних змін стінки тонкої кишки у різні терміни гострого порушення кровообігу в експерименті.

Abstract

Гостре порушення кровопостачання кишки супроводжується серйозним пошкодженням та руйнуванням слизової оболонки. Оскільки руйнування кишкової стінки можна спостерігати в цьому потенційно летальному стані, ми вивчили розвиток і послідовність подій, оцінюючи морфологічні зміни стінки тонкої кишки в моделі гострої ішемії в анестезованих щурів. Метою даного дослідження є детальний аналіз та систематизація комплексу морфологічних змін усіх шарів стінки тонкої кишки в експериментальній моделі гострого порушення кровообігу, з особливою увагою до послідовності та характеру деструктивних і регенеративних процесів у різні терміни ішемії. Морфологічні дослідження проводили в динаміці через 3, 6, 8 та 12 год з моменту перев’язки верхньої брижової артерії. При розтині, окрім зовнішнього огляду, забирали шматочки тканини тонкої кишки. Для гістологічного дослідження після фіксації та проведення за загальноприйнятими методами матеріал заливали в парафінові та целоїдинові блоки. Проведе це дослідження чітко демонструє, що час 3-6 год є критичним «вікном» для розвитку незворотного, життєзагрозливого пошкодження кишки, яке швидко переходить у системну катастрофу. Головною причиною летальності є повна втрата цілісності кишкового бар’єру та активний колапс мікроциркуляторного русла. Отримані дані слугують об’єктивною основою для розробки та верифікації нових методів фармакологічного захисту кишкової стінки, спрямованих на збереження мікроциркуляції та бар’єрної функції у ранні терміни ішемії. Acute impairment of intestinal blood supply is accompanied by severe damage and destruction of the mucous membrane. Since the destruction of the intestinal wall is observed in this potentially fatal condition, we studied the development and sequence of events by evaluating the morphological changes in the small intestine wall in an acute ischemia model in anesthetized rats. The objective of this study is the detailed analysis and systematization ofthe complex ofmorphological changes across all layers ofthe small intestine wall in an experimental model ofacute circulatory disturbance, with particular attention to the sequence and nature of destructive and regenerative processes at various time points ofischemia. Morphological studies were conducted dynamically at 3, 6. 8, and 12 hours after the ligation of the superior mesenteric artery branch. During the dissection, in addition to external examination, tissue samples from the small intestine were collected. For histological examination, the material was fixed, processed using standard methods, and embedded in paraffin and сelloidin blocks. The conducted study clearly demonstrates that the 3-6-hour period represents a critical «window» for the development ofirreversible, life-threatening intestinal damage that rapidly progressesto a systemic catastrophe. The main cause of mortality is the complete loss of intestinal barrier integrity and the active collapse ofthe microcirculalory bed. The obtained data serve as an objective basis for the development and verification of new methods of pharmacological protection ofthe intestinal wall, aimed at preserving microcirculation and barrier function in the early stages of ischemia.
